What is Systemic lupus erythematosus or SLE? Systemic lupus erythematosus or SLE is a multisystem autoimmune disease of protean manifestations and variable behavior. Clinically, it is an unpredictable, remitting and relapsing disease of acute or insidious onset that may involve virtually any organ in the body; however, it affects principally the skin, kidneys, serosal membranes, joints, and heart. The fundamental defect in SLE is a failure to maintain self-tolerance. One model for the pathogenesis of the disease proposes a combination of increased generation or defective clearance of nuclear antigens released from apoptotic cells, and a failure of T- and B-cell tolerance to these self-antigens
